The main character of Red Dead Revolver is Red Harlow, a bounty hunter voiced by Robert Bogue who hunts down the men who murdered his parents. Five other characters are playable for a chapter each: Jack Swift, Annie Stoakes, Shadow Wolf, General Diego and the Buffalo Soldier. The villains are General Diego, his right-hand man Colonel Daren and Governor Griffon.

Colonel Daren, working for General Diego, killed Red's parents, and Red kills them both. The final boss is Governor Griffon, revealed as Griff, the partner who betrayed Red's father Nate years earlier.

Annie Stoakes runs the ranch she inherited from her late father near Bear Mountain in Red Dead Revolver (2004). She refuses to sell her land to Governor Griffon, who has it burned, and becomes one of Red Harlow's allies. Her rifle is called Faith. Carrie Keranen voices her.

General Javier Diego is a former Mexican army general leading a renegade army in Red Dead Revolver (2004). He spared Griff in exchange for the Bear Mountain gold, sent Colonel Daren to kill Red Harlow's parents, and runs the mine with forced labour. Red kills him. Robert Jimenez voices him.

In the flashback chapter The Traitor, playable as Diego, the general blows a bridge and captures Griff, who offers him half of the gold mine to save his life, and Diego suggests killing his partner Nate. Diego then sends Colonel Daren to kill the Harlow family. Prisoners work the Bear Mountain mine, and Diego delivers its gold to the Governor.

Governor Griffon, once known as Griff, was Nate Harlow's partner in the Bear Mountain gold claim. In Red Dead Revolver (2004) he is the governor and the final boss: the man whose betrayal led to the murder of Red Harlow's parents. Red kills him in a rooftop duel. Bert Pence voices him.
Griff and Nate Harlow shared the Bear Mountain claim and a pair of identical revolvers. Captured by General Diego, Griff offered half of the gold and pointed to Nate as the owner of the other half, which led to the Harlow murders. More on General Diego.

Jack Swift is an English trick-shooter who fights alongside Red Harlow in Red Dead Revolver (2004). Red frees him from a cage at the circus of his former employer, Professor Perry, and Jack later joins the assault on Governor Griffon's mansion. He is playable in one chapter. Gregg Martin voices him.

Red Harlow is the protagonist of Red Dead Revolver (2004): a bounty hunter hunting the men who murdered his parents when he was a boy. Son of the prospector Nate Harlow and Falling Star, he takes his revenge on Colonel Daren, General Diego and Governor Griffon, and survives. Robert Bogue voices him, and Jason Fuchs voices him as a teenager.

Nate Harlow and his partner Griff found gold at Bear Mountain and had a pair of identical revolvers made. Captured by General Diego, Griff betrayed Nate to save himself, and Diego sent Colonel Daren to raid the Harlow home, where Nate and Falling Star were killed. Young Red shot off Daren's left arm with his father's revolver, picked up from the fire, and the red-hot gun left a scorpion-shaped scar on his hand.
