The main characters of Bully are Jimmy Hopkins, the 15-year-old protagonist voiced by Gerry Rosenthal; Gary Smith, the friend who becomes his enemy (Peter Vack); Pete Kowalski, his loyal friend (Matt Bush); and principal Dr. Crabblesnitch (Ralph Gunderman). The rest of the cast is split into six cliques, each with a leader Jimmy must beat: Russell, Derby, Johnny, Earnest, Ted and Edgar.

Bullworth's students are divided into Bullies, Preppies, Greasers, Nerds and Jocks, and the Townies are former students living in town. Jimmy takes over the cliques one by one, and each chapter ends with a fight against a leader.

Gary befriends Jimmy on day one, then betrays him by sending him to fight Russell. From then on, Gary works behind the scenes: he turns the Preppies against Jimmy, warns Johnny Vincent about Jimmy and Lola, and finally recruits the Townies to wreck the school and get Jimmy expelled. Derby Harrington is the leader of the Preppies, Bullworth's rich kids, in Bully (2006). After Jimmy beats Bif at boxing, Derby sets his clique on him and becomes the Chapter 2 boss. He is voiced by John Lavelle, spelled LaVelle by Bully Wiki and Behind The Voice Actors.

Dr. Crabblesnitch is the headmaster of Bullworth Academy in Bully (2006). He sees bullying as "school spirit", trusts Gary Smith, and expels Jimmy Hopkins before setting things right at the end. He was first planned as the game's main antagonist, a role that went to Gary. Ralph Gunderman voices him.

Crabblesnitch's office is on the second floor of the Main Building, up the stairs from the entrance, guarded by his secretary, Miss Danvers, voiced by Lori Funk. He teaches no class. You see him when you get busted: caught on campus during class time, Jimmy is taken to the principal's office and loses his weapons, except the slingshot, the skateboard and the camera. Get busted often enough and you land in detention, mowing the lawn or shoveling snow; completing every detention unlocks the prison uniform.

Ralph Gunderman was born on 6 September 1948 in Cleveland, Ohio. For more than a decade he was the announcer of NBC's Dateline, and he voiced the fake announcements in the monologues of the Late Show with David Letterman. He also did voices in Grand Theft Auto V and additional voices in Star Wars: The Old Republic. He retired in 2021 and died on 1 March 2026, aged 77, in New York.
Johnny Vincent is the hot-tempered leader of the Greasers in Bully (2006), jealous and devoted to his unfaithful girlfriend Lola Lombardi. Jimmy Hopkins works for him, fights him at the junkyard in Chapter 3, then frees him from an asylum in Chapter 5. Rocco Rosanio voices him.

Peter "Pete" Kowalski, nicknamed Petey, is one of Jimmy Hopkins' first two friends at Bullworth Academy in Bully (2006), with Gary Smith. Small, physically weak and picked on by Gary, he is also a sharp strategist who stays at Jimmy's side to the end and becomes head boy. Matt Bush voices him.

Zoe Taylor is a former Bullworth student who hangs out with the Townies when Jimmy Hopkins meets her in Chapter 5 of Bully (2006). Expelled after accusing the gym teacher Mr. Burton of harassment, she helps Jimmy win back the school, is readmitted and ends the game as his girlfriend. Molly Fox voices her.
Zoe reported Mr. Burton for sexual harassment, and she was the one expelled. At the end of the game, Burton is fired and Zoe is readmitted.
