The main characters of Manhunt 2 are Daniel Lamb, a former scientist voiced by Ptolemy Slocum, and Leo Kasper, voiced by Holter Graham, who helps him escape the Dixmor asylum. The twist is that Leo is not a real person: he is an assassin's personality implanted in Daniel's brain by a secret research program called the Project.

Leo seems to be a fellow inmate who leads Daniel out of Dixmor. In reality, the Project's Pickman Bridge, a brain implant designed to hold an assassin's personality, malfunctioned in Daniel, who perceives that personality as a separate person: Leo. Dr. Daniel Lamb is the protagonist of Manhunt 2 (2007): a former scientist of the Project, a government mind control program, held for six years in the Dixmor asylum. He escapes with Leo Kasper, recovers his memories, and learns that Leo is a personality implanted in his own mind. Ptolemy Slocum voices him.
Daniel studied biochemistry at the University of San Fierro, which places Manhunt 2 in the GTA universe. Married with two children, he volunteered to test the Project's Pickman Bridge implant to secure his family's future. The implant malfunctioned and gave him a second personality: Leo Kasper, a trained assassin, who calls him "Danny". More on Leo Kasper.

The game runs through 14 episodes: Awakening, Ghosts, Sexual Deviants, Red Light, Best Friends, Safe House, Bees' Honey Pot, Assassination, Most Wanted, Ritual Cleansing, Origins, Broadcast Interrupted, Altered State and Domestic Disturbance, then Personality Clash or Release Therapy. In some of them you play as Leo instead of Daniel; both handle the same. The flashbacks to 2001 show what Daniel's body did:

On consoles, Release Therapy unlocks once you have finished the game; its conditions have varied between versions.
Daniel kills through stealth executions in three levels, white, yellow and red, depending on how long you stay behind the enemy. He can also execute with a firearm, use the scenery (a phone or a manhole cover, marked by a skull on the radar) or drop onto enemies from above. On Wii, you finish the executions by copying on-screen gestures with the Wii Remote and Nunchuk. In the M-rated console release, the executions are blurred by color and motion filters.
Ptolemy Slocum gives Daniel both his voice and his motion capture. Born on 20 November 1975 in Nairobi, Kenya, he is best known as Sylvester in HBO's Westworld, and has appeared in The Wire, The Sopranos, Fringe and Veep. For Rockstar, he also voiced Steve, co-host of The Electron Zone on Liberty City Free Radio, in GTA Liberty City Stories (2005). In Manhunt 2, Michael Grant is voiced by Baron Vaughn, Doctor Pickman by Richmond Hoxie and Dr. Whyte by Nicole Orth-Pallavicini.
Leo Kasper is the antagonist of Manhunt 2 (2007). He first appears as Daniel Lamb's fellow inmate and guide out of the Dixmor asylum, but he is not a separate person: he is the assassin personality implanted in Daniel's mind by the Project's malfunctioning Pickman Bridge. Holter Graham voices him.
Only as a personality. The Pickman Bridge implant carries the personality and skills of a trained assassin, and its malfunction gave Daniel a split personality: Leo. More on Daniel Lamb.
Taking over Daniel's body, Leo killed Project members and police officers, burned the Project's records on Daniel and himself in Ritual Cleansing, killed Daniel's best friend Michael Grant, and killed Daniel's wife to weaken Daniel's hold and take full control.
Only in the alternate ending, Release Therapy, where Leo defeats Daniel and controls his body. In the main ending, Personality Clash, Daniel destroys him inside his own mind. After escaping Dixmor, Leo guides Daniel and pushes him to kill.
